Bullet Sizing Question

I am curious if anyone has any mathematical equation to start with on sizing conical bullets for each particular barrel? It seems the 50 caliber barrels I have measures are ~.500" across the lands but the groove depth varies. Is there anythig to it other than testing the fit and follow up shot loadability? (hunting).
